Nifurtimox
Mechanism :
Inhibits trypanothione reductase, a parasite-specific antioxidant defence enzyme.
Indication :
- American Trypanosomiasis (Orphan drug)
Contraindications :
Active or history of peripheral neuropathy, Active or history of seizures & cerebral impairment, such as behavioural disorders, epilepsy, or psychoses, Hepatic or renal impairment.
Dosing :
Oral:
1-10 years:
15-20 mg/kg/day every 8 hours.
11-16 years:
12.5-15 mg/kg/day every 8 hours.
Duration:
90 days.
Adverse Effect :
Anorexia, ataxia, convulsions, depression, fatigue, headache, memory impairment, paresthesia, sleep disorder, tremor, polyneuritis, weakness, rash, vomiting, myalgia, pulmonary infiltrates.
Interaction :
No known interactions.
Hepatic Dose :
Contraindicated in severe hepatic failure.
