Glycine
Mechanism :
Glycine is the major inhibitory neurotransmitter in the brainstem and spinal cord, where it participates in a variety of motor and sensory functions. It has recently been shown to function as a co-agonist at the N-methyl-D-aspartate (NMDA) subtype of glutamate receptor. In the latter, context glycine promotes the actions of glutamate, the major excitatory neurotransmitter. Thus, glycine subserves both, an excitatory and an inhibitory action.
Indication :
Contraindications :
Contraindicated in anuric patients and in liver/renal impairment.
Dosing :
50 mg/kg/dose TDS. Dose may be increased to 250 mg/kg/day in 3 divided doses during acute episodes (Maximum dose).
Adverse Effect :
None known.
Interaction :
Aspirin: Interferes with synthesis of isovaleryl glycine and is therefore contraindicated.
Hepatic Dose :
Use with contraindicated in patients with hepatic insufficiency because of the risk of developing or worsening neurological disorders.
