Loratadine

Mechanism :

Loratadine is a long-acting tricyclic antihistamine with selective peripheral histamine H1-receptor antagonistic activity.


Indication :

• Urticaria

• Allergic rhinitis


Contraindications :

Loratadine is contraindicated in patients who are hypersensitive to this medication or to any of its ingredients.


Dosing :

2-6 years: 5 mg, orally every day.

>6 years: 10 mg orally, every day.


Adverse Effect :

Nausea, headache, somnolence, drowsiness, nervousness, fatigue, dry mouth, hyperkinesia, conjunctivitis, dysphonia, malaise, abdominal pain, tachycardia, syncope, alopecia, anaphylaxis, abnormal hepatic function, supraventricular tachycardia.


Interaction :

Cimetidine, Erythromycin, Ketoconazole, Quinidine, Fluconazole, Fluoxetine: Could elevate plasma loratadine levels, which could theoretically result in adverse effects.

Hepatic Dose :

Children 6 years and older: Reduce initial dose to 10 mg PO every alternate day.
