Drotaverine

Mechanism :

Drotaverine inhibits phosphodiesterase hydrolysing cAMP, thereby increasing cAMP concentration, decreasing Ca uptake of the cells and changing the distribution of calcium among the cells. It may also have minor allosteric calcium channel blocking properties. It is a smooth muscle relaxant.


Indication :

  • Functional bowel disorders
  • Renal colic pain relief

Contraindications :

Hypersensitivity & contraindicated in patients with severe heart, liver, and kidney disease.


Dosing :

Oral
1-5 years:
20 mg, 3-4 times daily.
6-12 years:
40 mg thrice daily. Max dose: 80 mg/dose.

Adverse Effect :

Fainting, nausea, vomiting and dry mouth.


Hepatic Dose :

Use is contraindicated in severe hepatic impairment.
